B73 inbred line were cultivated using a hydroponic system under controlled conditions. For cadmium treatment, the trifoliate seedlings were transferred into fresh growing solutions containing 100 μM CdCl2. After 1 hr of cadmium treatment, maize seedlings of untreated and cadmium treated roots were collected for RNA-Seq analysis. Total RNA was extracted from maize seedling roots using a plant RNA kit (Omega, USA) following the manufacturer's instructions. RNA-seq reads were mapped by MaizeGDB to Zm-B73-REFERENCE-NAM-5.0. using STAR. Bigwig files were generated using bedtools genomecov -split -scale 1.0 followed by kentUtils bedGraphToBigWig.
