The seeds were then germinaged in the dark at room temperature. The seedlings were then transferred to plastic bags when roots were 1cm long. Seedlings with two visible leaves were then transferred to porcelain pots containing nutrient solution after the endosperm was removed. The plants were transfered to half strength of nutrient solution to full strength of nutrient solution (containing high nitrogen).The plants were grown in a growth chamber at 28/22 °C with a 14/10 h light/dark cycle with photosynthetic photon flux density at 250–300 µmol·m−2·s−1 at canopy height at the light cycle. Total RNA was isolated from mature tissue of expanding leaf samples. RNA-seq reads were mapped by MaizeGDB to Zm-B73-REFERENCE-NAM-5.0. using STAR. Bigwig files were generated using bedtools genomecov -split -scale 1.0 followed by kentUtils bedGraphToBigWig.
