Name: Variation in leaf transcriptome responses to elevated ozone corresponds with physiological sensitivity to ozone across maize inbred lines Link to publication: https://doi.org/10.1093/genetics/iyac080 NCBI bioproject ID: PRJNA604929 Category: RNA-Seq, stress Description: Seeds of maize genotype B73 were grown in LCI mix in growth chambers set to 15h day at 25oC and photosynthetic photon and photosynthetic photon flux of 350 µmol m-2 s-1 , 9 h night at 21oC and a relative humidity of 60%. The plants were fertilized (Osmocote Blend 19-05-8) at the start of the experiment. Control plants were grown at low ozone conditions (<10 ppb) or under ambient conditions and ozone treated plants were fumigaed with 100 pp ozone for 6 h per day from emergence to sampling. The ozone was generated using a variable output UV-C light bulb ballast (HVAC 560 generator, Crystal Air, Langley, Canada). leaf tissue from the fifth leaf is sampled for processing. Total RNA was isolated independently for each sample.RNA-seq reads were mapped by MaizeGDB to Zm-B73-REFERENCE-NAM-5.0. using STAR. Bigwig files were generated using bedtools genomecov -split -scale 1.0 followed by kentUtils bedGraphToBigWig.
