B73 was growm in a greenhouse under short-day condition at 12 h light and 12 h dark and temperatures of 26 to 28°C during day and 23–25°C at night. The plants were divided into two well watered and drought stress when the tassel was visible. Drought stress plants were maintained at 1.5 MPa (5–8% soil water content) and control plants at 0.2 MPa (15-20% soil water content). Leaf tissue was collected from the flag leaf of replicates of both drought and well watered plants. Total RNA was extracted from each sample using RNeasy Plant Mini Kit (Qiagen). RNA-seq reads were mapped by MaizeGDB to Zm-B73-REFERENCE-NAM-5.0. using STAR. Bigwig files were generated using bedtools genomecov -split -scale 1.0 followed by kentUtils bedGraphToBigWig.
